Effects of Nitric Oxide Produced by Macrophages on the Proliferation of Murine Lymphocytes Stimulated by Mitogen

Abstract

|
Nitric oxide(NO) has been identified recently as a multifunctional mediator. NO has been known to be produced by most cells of the body and influence on most cells of the body. Besides its functions as endothelium-derived relaxing factor, as a neurotransmitter and as an immune defence molecule, evidence is accumulating that NO partitcipates in inflammation and autoimmjune-mediated destructijon of the tissues. The released NO could be toxic to immune and inflammatory cells as well as microorganisms and tumor cells. Thus, NO might be regarded as both immune effector and regulatory molecules.
In this study, the authors intended to observe the effects of NO produced by macrophages on the proliferation of lymphocytes stimulated by phytohemagglutinin(PHA) or concanavalin A (Con A) in mice and rats. DMacrophages of the body were primed and activated to release NO by the such strong antigen stimulations as Bacillus Calmette et Guerin infection or ultraviolet irradiatio. When the mouse lymphocytes were stimulated with PHA or Con A and corcultured with the maerophages which were stimulated to produce NO by the treatment with ¥ã-interferon(¥ã-IFN) and lipopolysaccharide(LPS), the proliferative activity of the lymphlocytes was strongly reduced. The reduction of lymphlocyte prolliferation was reversed when the inhibitory of NO production was strongly reduced. The reductionculture medium of lymphocytes and marcrophages which had been stimulated by PHA, and ¥ã-IFN and LJPS, respectively. The above results strongly suggest that NO might function as immunoregulatory molecule by the inhibitory action on the lymphocyte proliferation as well as immune effector molecule by the harmful effect on the invading microorganisms and tumor cells.
